We offer various CRISPR injection services, such as gRNA, gRNA + donor, and gRNA + ssODN injections.
With this type of service, we will not review your CRISPR design nor evaluate your CRISPR constructs.

gRNA Injections

In its simplest form we inject gRNAs for creating knockouts (indels). We inject ~250 nosCas9 embryos.

Currently used CRISPR strains: nos-Cas9mSA (II, P40) and nos-Cas9mSA (III, P2).

gRNA + donor Injections

We further inject gRNA constructs together with a donor construct for generating various types of knockouts or knockins. Currently, we suggest injecting ~500 Cas9 embryos to likely get several independent insertions of the donor.

Our goal is to achieve success with the very first CRISPR injection, avoiding the need for re-injections resulting in time delays. For this reason we inject a relatively high number of embryos.

If the goal (knockout, knockin) is not achieved with the first injection, our use of high embryo numbers strongly indicates that the failure most likely originates in something entirely unrelated to the injection (such as design errors, gRNA issues, construct flaws…).

FAQs

Currently we mainly use two strains: nos-Cas9-mSA (P40, on 2nd) and nos-Cas9-mSA (P2, on 3rd chromosome). In these strains the Cas9 is fused to a monomeric streptavidin (cas9-mSA).

These Cas9-mSA strains are healthy and their offspring has a good survival rate.

We prefer sending individual plasmids so that we are flexible with the final concentrations. Additionally, we can react on potential DNA quality issues. However, both options are acceptable.

For CRISPR transgenesis please provide 30 μl or more of your gRNA construct(s) at a concentration of 500 ng/μl or higher. For donor constructs, we require an amount of 30 μl or more at a concentration of 1 μg/μl or higher.

If you want us to take care of the DNA you can just send us a small aliquot of miniprep DNA and we will prepare injection quality DNA in-house for CHF 30 per sample.

We provide no free re-injection for a failed CRISPR experiment when we are not in charge of the CRISPR design and the generation of the reagents. The reason for this is that in a CRISPR experiment there are many parameters that are relevant for its success such as the CRISPR design, selection and efficiency of gRNAs, cloning-related issues. When you send us the CRISPR reagents we cannot control for any of these parameters. In our experience the failure of a CRISPR experiment most likely depends on one of such parameters or a combination of them and generally not on the injection itself.
