DNA quality is essential for successful transgenesis. Injection quality DNA (e.g. Qiagen Midiprep DNA) would be optimal. If you cannot provide such quality DNA, you can send us DNA miniprep sample(s) of your construct and we will prepare injection quality DNA for an additional charge of 30 CHF/sample.

Site-specific

For site-specific transgenesis please provide 30 μl or more of your DNA construct at a concentration of 300 ng/μl or higher. We will dilute the DNA to an appropriate injection concentration.

CRISPR

For CRISPR transgenesis please provide 30 μl or more of your gRNA construct(s) at a concentration of 500 ng/μl or higher. For donor constructs, we require an amount of 30 μl or more at a concentration of 1 μg/μl or higher.

P-element

For P-element transgenesis please provide 30 μl or more of your DNA construct at a concentration of 1 μg/μl or higher.

FAQs

What DNA preparation kit do you recommend?

We highly recommend the Qiagen midi prep kit.

The DNA quality is a key determinant of injection success. Poor DNA quality will lower the survival rate of injected embryos and decrease transgenesis rates or make transgenesis impossible. All this makes injections unnecessarily troublesome, and causes additional labor and costs.

Note, very poor DNA quality can lead to a complete loss of all the injected embryos and thus results in unnecessary additional costs and delays.

If you are not sure about the DNA quality that you can generate you can just send us a small aliquot of miniprep DNA and we will prepare injection quality DNA in-house for CHF 30. This is our preferred option.

What is the best way to send DNA samples?

It is not necessary to use ice or dry ice to send your DNA sample. Within of Switzerland regular mail is fine. If you send us your DNA samples from elswhere we recommend to use courier services or equivalent, mainly to avoid both long transportation time and loss of the samples, which sometimes happens with regular mail.
